Over 40 genes linked to depression identified, says a study

Researchers have identified 44 genomic variants that have a significant association with depression, an advance that may lead to improved antidepressant medications. The study, published in the journal Nature Genetics, is the largest genome-wide association to date of genetic risk factors for major depression. Of these 44 loci, 30 are newly discovered while 14 had been identified in previous studies. In addition, the study identified 153 significant genes, and found that major depression shared six loci that are also associated with schizophrenia.

“This study is a game-changer,” said Patrick F Sullivan, a professor at the University of North Carolina (UNC) in the US. The results can be used for improved therapies targets of known antidepressant medications were enriched in the genetic findings, researchers said. The genetic basis of depression overlaps importantly with other psychiatric disorders like bipolar disorder and schizophrenia, they said. Intriguingly, the genetic basis of depressive disorder also overlaps with that for obesity and multiple measures of sleep quality, including daytime sleepiness, insomnia and tiredness.

“We show that we all carry genetic variants for depression, but those with a higher burden are more susceptible,” said Naomi Wray, Professorial Research Fellow at the University of Queensland in Australia.“We know that many life experiences also contribute to risk of depression, but identifying the genetic factors opens new doors for research into the biological drivers,” said Wray.

Meditation may lower anxiety, boost heart health

Just a single session of meditation can alleviate anxiety and boost heart health, a study has found.

Researchers from Michigan Technological University in the US found that that 60 minutes after meditating the 14 study participants showed lower resting heart rates and reduction in aortic pulsatile load – the amount of change in blood pressure between diastole and systole of each heartbeat multiplied by heart rate.

Additionally, shortly after meditating, and even one week later, the group reported anxiety levels were lower than pre-meditation levels.

It sounds like a late-night commercial: In just one hour you can reduce your anxiety levels and some heart health risk factors. But a recent study with 14 participants shows preliminary data that even a single session of meditation can have cardiovascular and psychological benefits for adults with mild to moderate anxiety.

“Even a single hour of meditation appears to reduce anxiety and some of the markers for cardiovascular risk,” said John Durocher, assistant professor at Michigan Technological University.

While its well-documented that meditation over the course of several weeks reduces anxiety, there have been few comprehensive research studies on the benefits of a single meditation session.

Researchers wanted to understand the effect of acute mindfulness on cognition and the cardiovascular system to improve how anti-anxiety therapies and interventions are designed.

They designed the mindfulness study to include three sessions. First, in an orientation session researchers measured anxiety and conducted cardiovascular testing by measuring heart rate variability, resting blood pressure and pulse wave analysis;

Then there was meditation session that included repetition of the cardiovascular testing plus the mindfulness meditation – 20 minutes introductory meditation, 30 minutes body scan and 10 minutes self-guided meditation – as well as repeating cardiovascular measurements immediately following meditation and 60 minutes after.

This was followed by a post-meditation anxiety test a week later.

During a body scan, the participant is asked to focus intensely on one part of the body at a time, beginning with the toes. By focusing on individual parts of the body, a person can train his or her mind to pivot from detailed attention to a wider awareness from one moment to the next.

“The point of a body scan is that if you can focus on one single part of your body, just your big toe, it can make it much easier for you to deal with something stressful in your life. You can learn to focus on one part of it rather than stressing about everything else in your life,” said Hannah Marti, a recent Michigan Tech graduate.

Ketamine nasal spray can fight depression, suicidal thoughts

A novel nasal spray of ketamine — often misused as a party drug – can rapidly treat symptoms of major depression, and suicidal thoughts, a study has found.

The study, published in The American Journal of Psychiatry, compared an intranasal formulation of esketamine, part of the ketamine molecule, to a placebo for rapid treatment of symptoms of major depression, including suicidality, among individuals at imminent suicide risk.

The researchers at the Yale School of Medicine in the US involved 68 participants randomly assigned to one of two groups – either receiving esketamine or placebo twice a week for four weeks alongside standard treatment with antidepressants.

They looked at effects at four hours after first treatment, at 24 hours and at 25 days. Researchers, including those from Janssen Research and Development, found a significant improvement in depression scores and decreased suicidal ideation in the esketamine group compared to the placebo group at four hours and at 24 hours.

The esketamine effects were not greater than the placebo at 25 days. The measurement of suicide risk took into consideration both the patient’s and clinician’s perspectives.

The results of the study support nasal spray esketamine as a possible effective rapid treatment for depressive symptoms in patients assessed to be at imminent risk for suicide, researchers said.

Esketamine could be an important treatment to bridge the gap that exists because of the delayed effect of most common antidepressants.

Most antidepressants take four to six weeks to become fully effective. The scientists caution that more research is needed on the potential for abuse of ketamine.
